Detection of endocrine disrupting chemicals in Danio rerio and Daphnia pulex: Step-one, behavioral screen

چکیده

Anthropogenic surface and ground water contamination by chemicals is a global problem, there an urgent need to develop tools identify elucidate biological effects. Contaminants of emerging concern (CECs) are not typically monitored or regulated those with known suspected endocrine disrupting potential have been termed (EDCs). Many CECs be neurotoxic (e.g., insecticides) many incompletely characterized. Behavioral responses can neuroactive properties, which relevant EDC mechanisms neuroendocrine disturbances). Two freshwater species, Daphnia pulex Danio rerio, were evaluated for swimming behavior alterations resulting from 24-hr exposure 9 CECs: triclosan, triclocarban, chlorpyrifos, dieldrin, 4-nonylphenol, bisphenol-A, atrazine, metformin, estrone. This the first step in development bioassay detecting estrogenic and/or anti-androgenic activity goal evaluate complex mixtures uncharacterized contaminants samples. The second step, described subsequent report, examines transcriptome following chemical exposure. Significant differences response sensitivity found across within species given this unique optical system. In concentration ranges studied, significant behavioral detected 6 D. 4 rerio. These results underscore utility effects sublethal concentrations before exploration transcriptomic detection.

Brain and gonadal aromatase as potential targets of endocrine disrupting chemicals in a model species, the zebrafish (Danio rerio).

Many chemicals in the aquatic environment are able to adversely affect in vitro brain and ovarian aromatase expression/activity. However, it remains to be determined if these substances elicit in vivo effect in fish. With the view to further understanding possible effects of endocrine disrupting chemicals (EDCs) on aromatase function, we first developed methods to measure brain and ovarian arom...

Endocrine disrupting chemicals

The idea that xenobiotic chemicals could inappropriately modlate the endocrine system thereby causing detrimental effects n wildlife and humans was first proposed 20 years ago at the atershed Wingspread Conference [1,2]. These endocrine disruptng chemicals (EDCs) have long been controversial, with a great eal of discussion raging in the scientific literature. Much of this ontroversy has centere...

Cryptorchidism and endocrine disrupting chemicals

Prospective clinical studies have suggested that the rate of congenital cryptorchidism has increased since the 1950s. It has been hypothesized that this may be related to environmental factors. Testicular descent occurs in two phases controlled by Leydig cell-derived hormones insulin-like peptide 3 (INSL3) and testosterone. Disorders in fetal androgen production/action or suppression of Insl3 a...

Endocrine-disrupting chemicals and male reproductive health.

Endocrine-disrupting chemicals are substances present in the environment that can interfere with normal hormonal balance and thus exert potentially adverse health effects on the human organism. Male reproductive system development and function may be susceptible to the effects of such environmental toxicants. Bisphenol A, phthalates and alkylphenols are important components of multiple products...

Endocrine disrupting chemicals and disease susceptibility.

Environmental chemicals have significant impacts on biological systems. Chemical exposures during early stages of development can disrupt normal patterns of development and thus dramatically alter disease susceptibility later in life. Endocrine disrupting chemicals (EDCs) interfere with the body's endocrine system and produce adverse developmental, reproductive, neurological, cardiovascular, me...
